Nano CBN

Also known as: Cannabinol, CBN, Nano CBN

Overview

Cannabinol (CBN) is a mildly psychoactive cannabinoid found in the cannabis plant, typically formed as a degradation product of tetrahydrocannabinol (THC). Nano CBN refers to CBN formulated using nanotechnology to improve its solubility and bioavailability. It is marketed primarily for sleep enhancement, neuroprotection, and potentially for pain relief or anti-inflammatory effects. The nano formulation aims to increase water solubility and bioavailability, potentially leading to faster onset and enhanced efficacy compared to traditional CBN. Early-stage clinical evidence exists, mostly from small-scale studies and observational reports. Systematic reviews on cannabinoids focus more broadly on CBD and THC rather than specifically on CBN or nano-CBN. Therefore, there is limited high-quality RCTs specifically on nano CBN, and evidence is mostly from small observational studies and preclinical research. No large-scale meta-analyses exclusively on nano CBN were found.

Benefits

A survey-based study on a water-soluble nano CBN supplement reported significant reductions in sleep disorder incidence and severity across multiple sleep parameters, such as latency to sleep and difficulty initiating sleep. The reported reductions in sleep disorder incidence and severity are substantial, indicating potentially clinically meaningful improvements. The nano CBN product reportedly induced rapid sleep onset and improved sleep quality shortly after administration. Preclinical data suggest potential antitumor activity and selective cytotoxicity of nanoliposomal cannabinoid formulations, though this is preliminary and not yet clinically validated. The sleep study involved individuals suffering from various sleep disorders, and benefits were reported broadly across these groups.

How it works

CBN acts as a partial agonist at cannabinoid receptors CB1 and CB2, with weaker psychoactive effects than THC. It may modulate sleep through interaction with the endocannabinoid system, influencing neurotransmitter release and sleep-wake regulation. Primarily, it has central nervous system effects related to sleep regulation and possibly anti-inflammatory pathways. Known molecular targets are CB1 and CB2 receptors, with potential indirect effects on GABAergic and glutamatergic neurotransmission. Nano formulations improve water solubility and bioavailability, leading to higher plasma concentrations and faster onset compared to conventional formulations.

Side effects

Nano CBN is generally well tolerated in small observational studies, with mild adverse effects that resolved with dose adjustment. Mild adverse effects were reported in a small percentage of individuals, but specifics were not detailed. No uncommon or rare side effects were reported in available studies. Due to cannabinoid interactions with cytochrome P450 enzymes, caution is advised regarding potential drug interactions. Contraindications are not well established, but caution is advised in pregnant or breastfeeding women, and those on CNS depressants. Data is lacking for special populations, so more research is needed.

Dosage

Minimum effective dose and optimal dosage ranges are not established from current data. The sleep study used a proprietary nano CBN formulation with unspecified dosing. Maximum safe dose is also not established. Nano CBN appears to act rapidly to induce sleep, so timing before bedtime is implied. Nano or water-soluble formulations improve bioavailability and onset. Nanoformulation enhances solubility and absorption compared to traditional CBN. There are no required cofactors reported.

FAQs

Is nano CBN safe?

Preliminary evidence suggests it is well tolerated with mild side effects that resolve with dose adjustment.

Does nano CBN improve sleep?

Small observational data indicate significant improvements in sleep onset and quality.

How quickly does it work?

Nano CBN reportedly induces rapid sleep onset.

Is nano CBN better than regular CBN?

Nanoformulations improve bioavailability and solubility, potentially enhancing efficacy.

Are there any drug interactions?

Data are limited; caution is advised.

The study reported a significant reduction in the incidence and severity of sleep disorders with nano CBN, and any mild side effects resolved after dose adjustment. However, the study was non-randomized, lacked a control group, had a small sample size, and relied on self-reported outcomes.
  • https://pubs.acs.org/doi/10.1021/acsomega.3c05371 – This preclinical experimental study examined nanoliposomes for controlled release of cannabinol. The study found that nanoliposomal CBD showed improved solubility, controlled release, and selective antitumor activity in cell lines. This is preliminary, preclinical evidence, and no human data is available.

The review found that nano and pro-nano formulations of cannabinoids increase bioavailability and reduce the time to peak plasma levels. The review focused on CBD, with limited data on CBN specifically, providing indirect evidence for the effects of nanoformulation on CBN.
