7-Methoxyisoflavone

Also known as: Methoxyisoflavone, 5-methyl-7-methoxyisoflavone, 7-isopropoxyisoflavone, ipriflavone, 7-Methoxyisoflavone

Overview

7-Methoxyisoflavone is a naturally occurring methoxylated isoflavone, a subclass of flavonoids, primarily found in soybeans and other leguminous plants. It is classified as a phytoestrogenic flavonoid and polyphenolic compound. Historically, it has been marketed as a sports supplement for its purported anabolic (muscle-building) effects, though human research has largely failed to support these claims. Preclinical studies suggest potential benefits in bone health, cognitive function, and particularly in mitigating inflammatory conditions like atopic dermatitis. Its mechanism of action involves modulating signaling pathways, specifically the MAPK-AP-1 pathway, to influence inflammation and immune responses. While generally considered safe at studied doses, high-quality human evidence for its efficacy, especially for muscle growth, remains limited.

Benefits

While 7-Methoxyisoflavone has been marketed for anabolic effects, human trials have not supported these claims. An 8-week randomized controlled trial in males using 800 mg/day showed no significant improvement in muscle performance or lean body mass compared to placebo. However, preclinical research indicates significant anti-inflammatory benefits. A 2022 study demonstrated that 7-Methoxyisoflavone effectively reduced symptoms of atopic dermatitis in mice by regulating Th1/Th2 chemokine balance and inhibiting MAPK-AP-1 signaling pathways, leading to a significant reduction in inflammatory cytokine production (e.g., Cxcl9, Cxcl10, Ccl17, Ccl22). This suggests a strong anti-inflammatory potential, though these findings require validation in human studies. Secondary effects such as potential bone mineral density improvement and cognitive benefits have been suggested in animal studies, but robust human data is currently lacking.

How it works

7-Methoxyisoflavone primarily exerts its effects by inhibiting the MAPK-AP-1 signaling pathway. This involves reducing the phosphorylation and activation of key kinases such as Ras, c-Raf, ERK, JNK, and p38. By suppressing these components, it subsequently inhibits the activation of AP-1 transcription factors, specifically c-Fos and c-Jun. This inhibition leads to a decrease in the production of various chemokines and cytokines, thereby modulating the immune response and balancing Th1/Th2 chemokine expression. As a phytoestrogen, it may also interact with hormone-sensitive tissues, although the extent of this interaction and its physiological significance are not fully elucidated.

Side effects

7-Methoxyisoflavone is generally considered safe, with no significant adverse effects reported in human trials at doses up to 800 mg/day. Common, uncommon, or rare side effects have not been documented in available research. However, due to its phytoestrogenic activity, there is a potential for interactions with estrogen-sensitive conditions. Caution is advised for individuals with estrogen-sensitive cancers or those at high risk for such conditions. Specific drug interactions have not been extensively studied, but individuals on hormone-related medications should consult a healthcare professional. Safety in pregnant or lactating women has not been established, and its use is not recommended in these populations.

Dosage

There is no established minimum effective dose for anabolic or anti-inflammatory effects in humans due to limited research. In studies investigating muscle performance, a dosage of 800 mg/day was used in resistance-trained males, but it showed no benefit. This dose appears safe in short-term studies, but a maximum safe dose has not been well-defined. There are no specific timing recommendations, as studies typically involved daily dosing. 7-Methoxyisoflavone is generally consumed as oral supplements in capsule or powder form. Its absorption is likely similar to other isoflavones, potentially influenced by gut microbiota. No specific cofactors are identified as required for its efficacy.

FAQs

Does 7-Methoxyisoflavone build muscle?

Current high-quality human evidence indicates that 7-Methoxyisoflavone does not significantly increase muscle mass or improve muscle performance.

Is 7-Methoxyisoflavone safe to use?

Yes, it is generally considered safe at studied doses (up to 800 mg/day) with no significant adverse effects reported in human trials.

Does 7-Methoxyisoflavone have anti-inflammatory effects?

Yes, preclinical animal studies show it reduces inflammation in conditions like atopic dermatitis by inhibiting the MAPK-AP-1 pathway.

Can 7-Methoxyisoflavone affect hormone-sensitive conditions?

Due to its phytoestrogenic properties, caution is warranted, especially for individuals with estrogen-sensitive conditions or cancers.

How long does it take to see effects from 7-Methoxyisoflavone?

No proven anabolic effects exist. Anti-inflammatory effects in animal models have been observed within days to weeks of administration.

Research Sources

  • https://www.nature.com/articles/s41598-022-12695-3 – This animal study demonstrated that 7-Methoxyisoflavone significantly alleviated symptoms of atopic dermatitis in mice. It achieved this by regulating Th1/Th2 chemokine balance and inhibiting the MAPK-AP-1 signaling pathway, leading to a reduction in inflammatory cytokine production. The research provides strong mechanistic evidence for its anti-inflammatory potential in a preclinical setting.
  • https://pmc.ncbi.nlm.nih.gov/articles/PMC3435905/ – This human randomized controlled trial investigated the effects of 800 mg/day of methoxyisoflavone supplementation over 8 weeks in resistance-trained males. The study concluded that 7-Methoxyisoflavone did not lead to significant improvements in muscle performance or lean body mass compared to a placebo group. No adverse effects were reported, indicating its safety at this dose, but its ineffectiveness for muscle building.
  • https://www.fisiologiadelejercicio.com/wp-content/uploads/2017/04/Polyphenols-and-Performance.pdf – This systematic review and meta-analysis examined the effects of various polyphenols on exercise performance. While it highlighted some performance benefits for other polyphenols like quercetin, 7-Methoxyisoflavone was not specifically included or supported as an effective compound for enhancing athletic performance in this broader context. This indicates a lack of robust evidence for its performance-enhancing claims.
