Methyliberine

Overview

Methylliberine, commonly known by its brand name Dynamine®, is a purine alkaloid often synthesized for use in dietary supplements. It is primarily used to enhance energy, focus, concentration, and mood. Similar to caffeine and theacrine, methylliberine acts as an adenosine receptor antagonist but has a shorter half-life. Research on methylliberine is relatively limited compared to more established stimulants, but available studies, including randomized controlled trials and within-subject crossover designs, suggest potential benefits. It is found to improve concentration, motivation, and mood without significantly affecting vital signs or cognitive function. Methylliberine reaches peak plasma concentrations at 0.8–0.9 hours and has a half-life of 1.4–1.5 hours.

Benefits

Methylliberine has been shown to improve concentration, motivation, and mood. A double-blind, randomized, within-subject crossover trial found that 100 mg of methylliberine significantly improved concentration at 1 and 3 hours, motivation at 3 hours, and mood at 1, 2, and 3 hours (p ≤ 0.050). Women in the study also experienced elevations in sustained energy at 1 and 3 hours compared to placebo (p ≤ 0.050). Additionally, methylliberine improved well-being and the ability to tolerate stress to a greater extent than the placebo at 3 hours. These benefits were observed in both men and women, with specific improvements in sustained energy noted in women. The effects are noticeable as early as 1 hour post-ingestion and can last up to 3 hours.

How it works

Methylliberine primarily functions as an adenosine receptor antagonist, which can increase dopamine transmission and impact behavioral activation and effort-related processes. By modulating adenosine receptors in the central nervous system, it leads to increased alertness and energy. Its primary molecular targets are adenosine A1 and A2 receptors. Methylliberine reaches peak plasma concentrations at 0.8–0.9 hours and has a half-life of 1.4–1.5 hours, indicating relatively rapid absorption and elimination from the body.

Side effects

Methylliberine appears to be safe when consumed in studied doses (25 mg and 100 mg). No common, uncommon, or rare side effects were reported in the reviewed studies. However, methylliberine can reduce the oral clearance and extend the half-life of caffeine when co-administered, although caffeine does not affect the pharmacokinetics of methylliberine. There are no specific contraindications identified in the available literature, and studies have not included special populations such as pregnant women, children, or individuals with chronic health conditions. More extensive and long-term studies are needed to fully understand its effects and potential interactions.
